anaerobic / microaerophilic, are they really different ? - (Nov/21/2008 )
Hi all
I am trying to growth a microaerophilic bacteria, however, I don't have any anaerobic jar or anaerobic gas pack.
how can I prepare 5 ml liquid culture easily ? without any anaerobic gas chamber ?
is that possible to make a burning candle inside the culture tube to remove oxygen ? (sound stupid....)

Have you tried just growing it in a mostly full tube with the lid on tightly? I've grown Borrelia, which is supposed to be microaerophilic and grown in a CO2 incubator, just in tubes with the lid on and they grow just fine. In fact, I think many Borrelia labs don't have CO2 incubators at all.
If you don't shake them you should be able to out a layer of mineral oil on top of it.
I grow 10 ml culture of S. pyogenes by closing the cap very tightly and that works perfectly. I only use anaerobic pouches or jars with gas pack for my plates. And I don't have CO2 incubator either.

I didn't try filling the tube full to remove most air inside.
However, should I remove dissolved oxygen in the solution too ? e.g. ultrasound or applying vacuum ?

you could sparge the solution with N2 gas filtered through .2 micron

actually, I am trying to grow my poor magnetic bacteria (M magnetotacticum), I tried all you guys suggestion, but it never growth.......
I followed the ATCC instruction but nothing happen....Is that bacteria really difficult to growth ???
