promoter cloning - (Nov/18/2008 )

Hi, i have a question regarding cloning of promoter.

I'm going to clone the promoter of my gene of interest into pgl3 vector for luciferase study. My question is, do I have to worry about whether the promoter is in frame and will case a frame shift?

I'm thinking that the answer is no, but i'm not sure why. blink.gif Will appreciate if someone can enlighten me with regards to this. Thanks alot.

Promoter has no frame. Frame is in the coding region (open reading frame). But when cloning, you need to make sure that

1. you are not introducing part of your gene ORF with your promoter
2. you are not disturbing the luciferase ORF.
