Is there any destructive effect on nucleic acid when harvest the cell by scrapin - (Nov/13/2008 )

Dear all,

I have to extract cell RNA from a T25 flask, and i'm going to first harvest the cell with cell scraper as part of the cells are required for protein extraction with RIPA lysis buffer . Will it cause any damage to the cell's RNA by scraping?

There shouldn't be much direct damage to the actual RNA, though you will burst/damage a large number of cells which may cause an up-regulation of some genes unless you are quick with your harvest.
