EDTA and EGTA into solution - EDTA and EGTA into solution (Nov/11/2008 )

What is the saturation point? Would it be impossible to make a 600 mM EGTA and 600 mM EDTA solution?

For 100 mL this would involve 22.9 g EGTA and 22.3 g EDTA in 100 ml and adjustment to ph8.

Id like to know if its possible to go that high, because Ive heard getting them into solution can be difficult.

Hey,

I assume that is related to your earlier questions about the calculations for your DNase inactivation.
So I think you can go higher than that. I would def. try a 1M solution. However, you have to add a little bit of NaOH. Otherwise you will not get it into solution.

You might have to heat it a little bit.

We routinely make up 500 mM EDTA, but be careful when adding your NaOH - it sometimes requires a lot depending on how you are adding it (5N, 10N, pellets). If you add too much water to begin with then your concentration will be off.

we use 1 M stock solutions of each, I think higher should also be possible; you have to add Tris or NaOH or KOH to solve EGTA or EDTA; mix extensively (hours!)
