temperature can effect the protein active? - (Oct/29/2008 )

I wan to express one protein in Ecoli, I select 27 degree 12 hours, 37 degree 4 hours and 16 degree 14 hours. who can tell me if there is different between them, and I found that different temperature, the amount of expressed protein was different. which situation is better for protein active, and I want to do in vitro test?

if iti s heterologue expression, it depends on species; a marine bacterium protein has other needs for temperature than mammalian cell protein
