How do I grow bacterial cultures in glycerol to saturation? I was thinking of leaving them at 37*C overnight gentle shaking and then they would be saturated the next day. Is this ok?

Sorry I was not specific enough. They are bacterial culture stocks stored in glycerol for many months. They are not at saturated level & I need to get them saturated for a certain experiment. How do I do that?
When you grow a culture from a glycerol stock, you have to inoculate a fresh solution of media (with ab if you need to). Generally, I slightly dip a pipett tip in the glycerol stock and I dump the tip in the tube containing the media. Cells will grow to saturation whithin the night (except if you have a slow growing strain).
Glycerol stock is, as its name implies, a stock. One tube can last for years. Do not waste it!
When waking up my bacteria cell, I streak my glycerol stocks onto selection plates.
I would place the frozen glycerol stock on dry ice. And scrap a bit of the stock using a wire loop and streak that loop on a plate. This keeps the stock from melting. you don't want to subject your frozen stock to multiple freeze thaw cycles. It is bad for the viability of the cells.
