Elimininating RNAses - (Oct/04/2008 )

Hi,

I am going to do Northern Blotting and it is important to make sure that my solutions are all RNase-free.
Is there any way to treat solution so that they will be RNase free, other than using DEPC?

A lot of times, you can buy the reagents already RNAse-free; look for molecular grade water, and mix reagents with that; you can also use RNAse-Away to treat/rinse any glassware or plasticware that youwill be using. In addition, make sure that you wear gloves when handling eppendorf tubes or whenever you fill up pipettor tip boxes, since the sutoclave doesn't kill RNAse.

Autoclaved millipore water is good for Northern blot. Wash your pipettes with ethanol before use ( the exterior only) and wash yourbench top too, it's never bad. Be sure your gel glasses are clean and make your gel mix ( if you don't buy them) in sterile container , like a Falcon tube.

Prepare your samples on ice.

That's about what I can think of now.
