Hi,

Can anyone tell me how to block endogenous peroxidase activity?
I never had any problem until i used sample conatins blood. I belive erythrocyte is somehow causing endogenous peroxidase activity.
Is there any way i can specifically block only the endogenous activity? i use caesin and Tween as my blocking agent works excellent in reducing background.

I would really appreciate your answers.

Hi,
Try 3% H2O2 for 10 minutes. Than rinse in PBS for 2 minutes. I do it in immunocytochemistry, before incubation with diluted serum.

I have had good results using 0.3% Peroxide in 0.1% sodium azide for 12-15 minutes.
