Problems with my THP-1 - (Sep/29/2008 )

Hello All,

I am new to cell line culturing and I have been told to maintain cultures of THP-1. Everything was fine until I revived a new batch of cells and saw that the cells are clumping? Why is this? what can I do to remove the clumping? I have some experience with cell cultures but mainly with HUVECs.

Cells are frozen in 90%FBS and 10% DMSO.

Cultures are maintained with RPMI supplemented with 10% Heat inactivated FBS and 2% Pen/Strep.

hay jackie...by 'clumping' do yuo exactly mean that they appear in dense clusters?...'coz THP-1 typically grows in grape bunch-like clusters...as long as its proliferating in number its quite normal...

Agree, macrophages grow like clumps. There should be more and bigger clumps in your flask when you culture them.
