Paraffin embedding - (Sep/28/2008 )

Hi,

Would like to have some explanation regarding about the paraffin embedding process before microtoming. Does the paraffin soak through the tissue itself, ie the paraffin diffuse throughout the tissue?

If so, is this the main reason why we could not labelled the tissue with fluorescence dye?

If you are using paraffin sections, you have to deparaffinize the sections before staining them. And it is possible to use fluorescent labelling on such sections. And when you mount them, you donot have to dehydrate them and mount using water based mounting media not solvent based.
