I take it you know how to count cells?

So, seed a several wells of a plate with cells at a known density (try 10,000 cells/cm2, and then lift and count the number of cells per volume of harvest over a set time course (e.g. every 2 days harvest a well), work out how many cells you have in total and plot those numbers on a time vs cell number graph. Exponential phase is where cell numbers are doubling or more per unit time, plotting on a log-linear scale will help identify it as it will become straight rather than curved.

Calculation for what? If for the curve, try some regression analysis, or get a graphing program (not Excel, it is terrible) and use that to tell you.
