Maturation of monocytes - (Sep/17/2008 )

Dear All,

I'm realtively new to the immunology field, so I was wondering if someone could help me. I am stimulating my monocyte cell line with LPS and looking at IL-1 beta production. However I'm getting strange results, so I'm wondering if I need to differentiate my non-transformed monocyte cell line, as maybe because they're isolated from the spleen they're still immature and won't respond properly? Is this could be the case, is there any way to maturate/differentiate them into mature monocytes or macrophages?

hi,
i think PMA is a strong stimulator over LPS (not sure 100%). try with 10ng/ml of PMA.

atleast you should get some signal of IL-1 upon stimulation as you have good percentage of fully transformed monocytes.
