pH lysis buffer - (Sep/15/2008 )

Hello!

Is there any specific reason why lysis buffers usually have a pH = 7.5? Since I load my protein extracts on SDS gels, would it be possible to lyse the cells with Tris pH = 6.8? :-)

it is near intracellular cytosolic pH, and prevents pH-dependent denaturizing during lysis for most proteins...
