Inhibitors in cell culture... - DMSO???? (Sep/10/2008 )
Hey,
I have an inhibitor to use on my cells.... and on the data sheet it says disolvable to 1mM in DMSO and 100mM in ethanol. Im disolving it to 1mM as a stick and then making my working dilution from there, but how do I know how much of the 1mM in 2 ml needs to be DMSO?? I know that the final conc of DMSO must be less than 0.1%.. so should I just work backwards or is there a way of knowing how much DMSO vs Water to use??

The entire 1mM has to be in DMSO. And make a 1000x or 500x stock solution if you are planning to add to cells.

Will change my first stock to a 5mM and then it'll be a 1:500 dilution. One last question...sorry.... Whihc is better for cells to have the inhibitor in DMSO or Ethanol? as if I increase my starting stock concentration I will have to use ethanol instead as DMSO is only soluble to 1mM not 5mM.

some DMSO-prediluted samples are still insoluble in water, so your inhibitor may precipitate after adding to cells...

I have tried both for cells and as long as you make up a 1000x or 500x stock, both of them are fine. I havn't used higher conc. of DMSO or ethanol.
