plasma membrane tag - (Sep/09/2008 )

Hi!

Could anybody suggest me a plasma membrane targeting signal. I would like to make membrane targeted fluorescent proteins. I tried the farnesylation motif of H-Ras in the C-terminus of my fluorescent protein, but still get localization in the ER(?) as well as other parts of cytoplasm. I would like to achive strong fluorescence only at the cell membrane. I have searched a lot in the internet, but cannot find any alternatives. Any ideas???

How about using membrane dyes like BODIPY? They are supposed to label all membranes.

How about using some sequences from caveolin or flotilin proteins?

Mistic, from B subtilis, targets proteins to the membrane. It's short, only ~100 residues, and also increases solubility.
