suspension cells - getting them to settle (Sep/03/2008 )

Hi,

Does anyone use suspension cells? i'm not used to using them but will be soon. at some point in my work i'm going to want to stick them on a culture slide. is there a method for getting suspension cells to stick to a slide?

Why do you need the suspension cells to stick to a culture slide?

I assume you want to do immuno on the cells at some point. This could be a problem with suspension cells. Most one's I have used are purely suspension cultures and will not normally stick to plastic. However you can try and use attachment factors such as:-
Polylysine
Fibronectin
Collagen
Primaria
Laminin
Gelatin


You could also try another immuno approach and label cells and use FACS analysis instead

it is indeed so we can do some immuno at a later point. we were thinking of putting the cells in serum free media. is it possible that would work, as have heard it might. not too keen to go down the facs route.

we've got suspension cells in serum-free media... seems to be fine.
to put onto a slide- poly-L-lysine coating seems to work out for us

HI,
we culture cells on serum-free media with-out problems...
and we put them on slices and seems to growth fine, but what kind of cells are you using??? We culture them on coverslips up to confluence and then, after a passage, drop an aliquot on the slice (and put the rest on new coverslip)
