what is difference of E.coli grow on different media? - (Aug/22/2008 )
Hi, Everyone:
I try to do some experiment on surface. At first, i use E.coli on the agar plate, then I heard that I should grow E.coli in LB broth, though I prepare solution of OD= 0.2. but both of E.coli gave different responce. Could someone tell me the difference of E.coli grown on these two different media? I am sure not kill the cell or break them during processing

What agar? LB?
Your differences might not be related to the composition of specific medium/agar, but rather to the conditions of growth.
which two media are you comparing?
But yes, different concentrations of nutrient (amino acids), types of carbon sources, nitrogen sources, oxygen availability will change the both the characteristics of E coli. This includes growth, exogenous protein expression, plasmid yields, etcs..
We don't know what "response" this guy thinks he's observing.

Sorry for discribing unclearly.
I used normal agar plate to grow e. coli. and take some colonies to dissolve in buffer solution. My reagent showed very positive responce to the existance of E.coli. However, I try to grow E.coli in LB broth, centrifuge, wash to get clean E.coli, then prepare the same buffer solution. My reagnt show no responce to the existance of E.coli. I think maybe during the growth, something appears on the surface of colony. if I use second method to grow E.coli, nothing special to give positive responce. Does someone know what appears during growing on agar plate but not growth in LB broth.
