IL4 and GMSCF for DC culture - how much IL4 and GMSCF for culturing dendritic cells (Aug/20/2008 )

I have searched for many papers and different protocols, and i find that the concentration of IL4 and GMSCF for culturing Dendritic cells are always vary greatly, i have found that people using different concentration for IL4 in DC culture, some are 10ng/ml, some are 20ng/ml, 50ng/ml some even use up to 100ng/ml. same case is also applied to GMSCF

So...can anyone share your concentration for culturing DC? and i got confused, in fact what concentration of IL4 and GMSCF should we use for DC culture?

what kind of DC?
human or mouse?
I use only 20ng/ml rmGMCSF(#315-03 peprotech) for murine bone marrow derived DC
it's OK.
If human, people say 1000U/ml GMCSF and 500U/ml IL4
you can use this formula:
UNITS/ml=1e6/ED50(ng/ml) to transform
tongue.gif

better is to titrate each cytokine in your own system...
check for cell death and maturation capabilities in various conditions
