Syber green - (Aug/11/2008 )

Hi mates,

iI have a syber green stock with 1000x, if i want to prepare the working solution which is 1x , with DMSO, did anybody know what is the DMSO concentration that i should use.

So if you're trying to get from 1000x to 1x then you need to dilute cyber green 1000 fold. This would be 1 ul of cyber green in 1 mL (1000 ul) of DMSO. I don't use cyber green so I assume you are asking for the correct dilution.

Starting from a 1000x SYBR green stock I prepared a 10x stock by adding 10ul SYBR (1000x) + 10ul DMSO (100%) + 980ul H20.
Once you dilute this 1:10 into your PCR mix (which is what I'm assuming you'll use it for) SYBR will be 1x and DMSO 0.1%.
If your PCR mix already contains some DMSO this should not affect much, on the contrary if it doesnt you might need to do some optimisation.

Thank you guys for the help, but i think i have checked that the working solution for syber green is 1 x but in this case you will have a working solution with 10x and a final conc will be 1x.

and is it enough to add just 10ul DMSO 100% to the 990 water+ Syber green?

He did give you a working 1x solution. It is just easier to work with if you make a 10x stock solution beforehand, and your results should be more consistent that way.
