DNA extinction coefficients - (Sep/29/2004 )

Hi,everyone,

I just want to use spectrophotometer to measure the concentration of my DNA. But I don't know the DNA extinction coefficients.
Does anyone know how to calculate the extinction coefficients and any website I can calculate online?

1 A260nm of ds DNA = 50µg/ml H2O
1 A260nm of ss DNA = 33µg/ml H2O

Pure DNA : A260nm/A280nm >= 1.8

1 A260nm of ss RNA = 40µg/ml H2O

Pure RNA : A260nm/A280nm >= 2.0

mind that ODs should lie between 0.1 and 1.0 for correct measurements!
