checking mRNA expression by reverse transcriptase PCR - (Aug/07/2008 )


I wanted to confirm the expression of my transgene by RT-PCR, I have designed the primers which will amplify ~250 fragment in its cDNA.

Can RNA be isoalted from a tail biopsy (mouse) so that cDNA can be generated to do RT-PCR ?

or a mouse has to be sacrificed to get some cells and then isolate RNA?

Can any one suggest me a protocol or a kit to isolate RNA and then do RT-PCR?

RNA can be isolated from any cells that make it, but you have to ask yourself which cells will be producing your transgene. Will the promoter you are using be functional in the tissues found in the tail biopsy?

My promter will not express the transgene in tail and so i have to sacrifice the mouse and take the right tisue.

can you suggest any reagent or kit to isolate RNA from eye lens?
