How to make my protein Soluble? - Bacterial system it sucksss ? (Jul/17/2008 )

I am handling a protein know to be secretary in normal. When i cloned With his tag, GST... it always goes into the pellet (Inclusion Bodies) .. I changed the temp, Strain,IPTG ..but no hope..... Kindly suggest me what else i can do to get my protein soluble..(In Bacterial System).

1. Can you express it without any tag? If so, you should be able to purify it from the medium, if it is, in fact, secreted.
2. There are a number of small tags that aid solubility, like mistic from B. subtilis, and Strep-Tag. I read once that making a double Strep-Tag increases solubility even further.
3. Can you purify and refold from the IB? Go to http://refold.med.monash.edu.au to look for tips on refolding IB proteins.
