Solubility of Oligonucleotides? - (Jul/07/2008 )

I have some oligonucleotides that I want to inject into the ventricles of the brain. I was going to use an azlet pump, ut now think, in our case, it would be better to just do daily injections. However, that means a MUCH more concentrated sample. 50-100 mg/ml type concentrations (as opposed to 2.5mg/ml for the pump).

Is such dilution of the oligos (from invitrogen and modified- phosphorothioate) a problem?

What's the solubility of oligos?

you may directly contact the manufacturer of the olgonucleotides, but i would see 2 problems :
I don't think it's well dissolved at this concentration and what will be the volume injected ? I mean if you want to make daily injections, is it possible to get a decent volume to inject at such concentration of the oligos ?

Well the volume is the problem. I can only inject so much volume at one time (as opposed to spreading it out over 24 hours, we are injecting over 5 minutes). So if I only get 5 minutes to inject, then I need a much more concentrated sample to inject that small volume containing the same amount of oligos. Any ideas?
