I need to refold protein and i want to do it using a urea gradient but i would like to know how long it takes for a full dialysis? thanks

I came to ask the exact same question about dialysis. Not for urea gradient, but to remove acid from cellulose extracted from paper. Currently, in our lab, we have some dialysis tubes which have been in dialysis for a month. I feel this is too long, but nobody actually listens when I say so. How long would you dialyse approx. 100ml for?
well i did bath of different concentrations at 4° for 3-5h as a minimum. Protein sample was in 2ml and buffer was 500ml. Then, i guess the overall duration of procedure depends of the nature of your protein. 1 month seems long to me too, but i guess there should be a reason for that (maybe forgotten )
Is the dialysis bag supposed to swell up with buffer? There were a few bubbles when i put it in the bath and 16 hr later, the bubbles are still there. Any thoughts or is this normal. Thanks
yes, it is normal for the bag to swell a little when dialyzing against a less concentrated solution. you can keep the swelling to a minimum by making the bag tight in the first place.

Isnt the tube supposed to fill up or will it stay the same, it was tight but i didnt want to lose any so i made it a little looser. The bag is supposed to take in buffer correct? Also can you dialzye with glycerol and/or triton x-100?
the tube will allow buffer to enter and increase volume until it is tight. any air bubbles, however, will remain. they won't pass through the membrane. in fact, we use that property to make the tube float in the solution.
glycerol and triton will pass through the membrane. in fact, anything small enough to fit through the pores can be dialyzed out or in.
