Non-directional cloning - (Jun/29/2008 )
Dear All,
I had done non-directional cloning for 5 different DNA samples obtained from different colonies from the same plate. However I detected the right size of the band only with 1 of those samples and rest of the other 4 samples does not seem to give the right size of the band. The other 4 have the same size .Can any senior enlighten me why does this happen in non-directional cloning?

Is there anything strange about the size? How does it compare with the 1 that was correct?
How pure is your insert? Remember that smaller fragments will tend to clone more efficiently than large inserts, and you might not necessarity be able to see them on a gel, if you're just doing an analytical gel.
