Elisa- need help - Elisa - Phosphorylation studies (Jun/27/2008 )

I am trying to set up an Elisa to quatitate ToTAL and Phosphorylated ERK1/ERK2 in tumor Lystes(not cell lysares)

I have purchades tke kit from R& D system. can anyone suggest a good tumor lysis buffer that would not affect the phosphorylation?
