northern with riboprobe - (Jun/28/2001 )

Hi! I made a riboprobe and now I would like to hybridise it on a northern. Maybe it is better if I load mRNA, but how much of mRNA I have to load on gel?Any suggestions are appreciatedmany thanksMonica

If you're doing total RNA extraction you should load about 10-15micrograms. If I remember correctly less than 1% of that is mRNA.Have you tried making a DNA probe. It definitely simplify things a lot. A riboprobe is more sensitive than a DNA probe is very liable to degradation.
