complete bisulfite treatment - How do you know if your DNA is completely modified by bisulfite treatm (Jun/23/2008 )

Hi,

I am thinking about how to confirm that my DNA sample is completely modified by bisulfite treatment.

I found two ways of this during search.

1. PCR with wild type primer(that is complementary to unmodified DNA) using the modified DNA as a template
2. PCR with methyl-specific primer using a universally unmethylated DNA

Which is the better way?

Thank you. Then, is any region in genome OK if it satisfies the criteria above?

I want to design the primers against areas without CpGs and with C's.
It would be very appreciated if anyone can help me. I am a newer in this field..
