PBS and STE Buffer - (Jun/18/2008 )

Hi All,
I`ve got a question and i need it be answered, what is the difference between PBS and STE Buffer. Am working now in tissue DNA Extraction, some protocols says wash the tissue with 1x STE buffer, other mention use PBS buffer, and am really confused. Acually, its my first time dealing with tissue samples so any suggestion from you would be higly appreciated.

Hi,

STE buffer is suitable for biomolecular lab procedures.1X solution contains 10mM Tris-HCl, 1mM EDTA and 100mM NaCl.Autoclaved.

1x PBS

8 g NaCl
0.2 g KCl
1.44 g Na2HPO4
0.24 g KH2PO4

in 800 ml of distilled H2O.

Adjust the pH to 7.4 with HCl. Add H2O to 1 liter. Sterilize by autoclave.

Thanx Sonken for the reply, my concern is which one is preferable for washing tissue samples prior to subjecting them to DNA extraction protocol, as i`ve mentioned earlier i reviewed some protocols but am not quiet sure which one is the best for such analysis.
