MOPS vs HEPES - which one do I use? (Jun/02/2008 )

Hi,

I am setting up a kinase assay for cdk5. After a quick lit search, I have found that some publications refer to a MOPS buffer, and some to a HEPES buffer for similar assays.

Now...because they have a similar pKa / similar properties - is it ok to just use either one?

I understand the ideal method would be to test the assay using both buffers - but i want a shortcut!

I would go with the HEPES because it is also used for tissue culture. Thus, your enzyme should be more stable in this environment.
