Hello....

I would like to know if TOP10F' or JM109 are methylase negative strains...

If I transform this cells with methylated plasmid, can I get un-methylated plasmid?

thanks....

welcome!

JM109 and TOP10F' are not methylation deficient strains, DNA in these cells will be methylated (check http://openwetware.org/wiki/E._coli_genotypes for E coli genotypes)
we normally use the INV110 strain from invitrogen to obtain non-methylated DNA
