Co-Transfection siRNA and targeting construct - (May/19/2008 )

Hi there,

my question is: Has anbody tried use siRNA and a targeting construct together for transfection? We did a control experiment where we transfected with pEGFP and siRNA and the targeting efficiency was quite low (around 20%, nomally we see 90% with pEGFG alone). So this I would also expect for the transfection with siRNA and the targeting construct, which will be used for stable transfection.
Do you have an idea how to enhance the targeting efficiency? Has anybody tied to do cotransfection?

cotransfection is routinely done. what is "the targeting efficiency"? did you mean % of specific knockdown of the target gene expression? How about change the ratio between siRNA and the target construct?

If you mean siRNA oligos and RNAi vector, it may improve the knockdown efficiency, but the transfection efficiency for oligos and vectors are different, they require different transfection reagents generally for max transfection. Also, there will be background effects doubling..

I do co-transfection of targeting construct with shRNA vector. It works well- it depends greatly on the ratio of the two plasmids or the target:siRNA ratio. You will really need to standardize that.
