count cell number - (May/14/2008 )

Hy boys!
How can i count the number of EUKARYOTIC cells in colture, using a spectrophotometer setted to 230, 260, 280, 320, 595 and 600 nm?
Please help me...

Generally you just use hemocytometer, but in batch cultures, people do spectrophotometry. The OD is 650nm.

As cellcounter said, people generally use a hemocytometer to count cells as part of passaging the cell lines. If you're trying to quantify the number of cells at the end of an assay, MTT is a common method. It really depends on what you're trying to count in what situations.
