The Best way of merging phase-contrast and fluorescence images of cells - in Photoshop (May/03/2008 )
Hello,
What is the best setting for merging a phase-contrast images and a GFP image of cells in photoshop. I know I need to have the two images on two layers, but what is the best blending mode and which layer should at top?

I dont know what software you are using. We have the software that comes with the scope and there is no different which one is on top of the other. You need to have right exposure for both.

I would not use photoshop for this, there are plenty of caveats, but basically, photoshop is not a program designed for science imaging (it can alter multiple parameters in non-lineal ways). You are best off using ImageJ and playing around with image calculator (add image) and altering brightness and contrast to the whole image. Referees can´t fault this but you should stick to clear guidelines (see J. Cell Biol What´s in a picture? 166(1) 11-15).
