Co-transformation - (Apr/26/2008 )

Hi all,

Quick question. I am currently trying to co-transform two plasmids (pET 24a and pACYC177 that has been modified for expression (insertion of a T7 promoter)), but when I try to isolate plasmid I get a 1Kb piece of DNA. Has anyone experienced this?

The cell line is an E. coli strain (delta ace, adhC, DE3)

DE3 is an protein expression line, they have endonucleases which digest the plasmid after 2-3 generations. If you want to grow and isolate the plasmid I would use an endA- line such as DH5alpha.
