genome-walker - (Feb/08/2002 )

Dear all,
i'm using a kit called "genome-walker" to "walk" upstream of my target gene. interestingly, i have got products with all the same sizes.... but i'm using different libraries to "walk" ... why do i get such a result?
pls help me ~~~

Most likely, this was caused by comtamination. the sources could be the primers, the templates or other reagents. I did some genome-walking before and only had this problem occasionally when I tried to perform a nested pcr.

But, after i have changed all my PCR buffer, MgOAc2, the water used, the primers also.. (fr the stock)
using all things new, even worse!!
i have got nothing!! even for the +ve control...
only smear, and smear for both first round & nested PCR... sigh~!

Which kit are you using? Is that from Clontech?

Dear Miffy

I have used two such kits from clontech and have never had such problem at least with the first round pcr. I am sure something definitely wrong with your system. You may want to check out with the company.
