Cell Culture Question - (Apr/10/2008 )
Why the FTS (Fetal serum albumin) is used for the culturing the cell.
and Cells are washed with the PBS why.
What things it removes from the culture.
Trypsin is also used for the enzymatic cleavage of the cell with the surface, does normal tapping can not be used to do that.
Fetal bovine serum provides proteins that cells need to proliferate. We add the FBS so the cells don't need to manufacture the proteins themselves, thus making their lives easier.
Cells are washed with PBS because you are removing all traces of used medium and dead cells from your culture. 1X PBS is an iostonic solution, so your cells are not damaged by the washing.
Not all cells will come off with normal tapping--some adherent cell lines need to be trypsinized to remove them from the culture surface. Other cells will come off in 1X PBS + 2mM EDTA.
I hope I have answered your questions.

Adherent cultures need to be washed with CaCl(-) and MgCl(-) PBS before being subcultured to remove the divalent cations that adhesion proteins need to bind, and to remove the FBS that will otherwise inactivate the Trypsin.
