analysing real Time data - Induction Ver percentage (Mar/24/2008 )

hi guys,

I have a problem with my final analysis of the data. I'm testing cytokines expression in control and treated cells taken form a WT and KO animals.

the effect I'm getting is quite clear about 50% difference between treated WT to KO, But the fold induction in each experiment is different (though the relative change stays the same, see blow in the example). therefore I can't show the mean Value (the SDTV are huge !!) or do a STAT test. (I can show the percentage of expression but than i can't do a t-test because the results of the WT (i.e. 100%) do not have a normal distribution pattern).

What can i do ?????

please help !!!!!

L.

example :

..........WT..... |....WT.............|... KO....|...KO
..........Ctrl.....|.....treatment...|...Ctrl....|...treatment
-------------------------------------------------------------
1)......... 1 ....|...1000............|... 1.......|.... 500
2)......... 1 ....|...8000............|... 1.......|.... 4000
3)......... 1 ....|...600..............|... 1.......|.... 300
4)......... 1 ....|...12000..........|... 1........|....6000

AFAIK you don't need any of that. If I understand it right and 1) to 4) are experimental replicates (cultivated at the same time with same conditions), so just say the relative ratio is always 50 fold (or make a mean of the relative ratios, to be precise) in 4 experiment replikates.
Because you were comparing them only within the number group not all to all.
Just count with the relative ratios if you need some statistics, others are from the nature of experiment uncomparable.
