primary hepatocytes as hepatocarcinogenesis model - (Mar/24/2008 )
plz suggest in vitro hepatocarcinogenesis model.
Can primary hepatocytes be used for evaluating the carcinogenic potential of various chemicals? I am confused because to culture primary hepatocytes in in vitro, one has to add many growth factors and these growth factors may have modulatory effect on the effects induced by carcinogen.am i right? if yes then, are there normal rat liver cell lines available for in vitro assessment of carcinogenic potential? plz help

growth factors at normal concentration for proliferation should not transform cells; if you have the opportunity to use primary hepatocytes, you should use it; some use immortalized hepatocytes which are an alternative

first of all thanks for your response. so in ur view i can use primary hepatocytes as hepatocarcinogenesis model... do u have any experience of culturing primary hepatocytes?

years ago, I did high pressure perfusion to isolate hepatocytes, a classic protocol and not easy...
