Hi, has anyone done siRNA transfection in suspension cells (Jurkat A3 cells)?
Also, which transfection reagent would you recommend for HEK 293 cells?

When i tried transfecting in suspension , i had more than 50% cells die so i don't know how good it is .
Hek293 should be possible with different reagents.
I would recommend HiPerFect from Qiagen.
They further offer protocols for suspension cells (maybe even for Jurkat cells).
I consider it to be one of the best reagents I have worked with.

I did some transfection with Lipo2000 a few years ago and it was hard
I got like 10-20% efficiency and end up needing to select for stably transfected cells.
Transient transfection of suspension cells is no easy job
I used Lipofectamine2000 to transfect HEK293 cells with siRNA for few years ago and it was quite efficient. Are you sure that your cells were okay?
For Jurkat cells, if I am not wrong you could try electroporation (Amaxa system).
try electroporation if you have one avaiable. suspension cells are tough to get siRNA in with most chemical methods.

sorry, I mean I used another cell line not HEK293, but also suspension cells.
Most adherent cells I transfected are very good, that's why I have an impression that transfection with suspension cell using lipo method is not easy
