inducible retro/lentiviral vector - (Mar/12/2008 )

Hi,

I need to express an apoptotic protein in primary macrophage therefore i need an inducible retroviral or lentiviral vector.

since i can't do a long selection i need a vector with either GFP marker or Puro selection.

Did anyone worked with that kind of vector ?

I have used an inducible retroviral system before. I fused my gene of interest with the estrogen receptor. In the absence of hormone (beta-estradiol), the gene is not expressed. Have a look on pubmed for Pax5ER or something similiar to get an idea smile.gif

the problem is that the type of macrophages i work with are quite responsive to estrogen .... i need something else (like a tet on system).

You may want to look over the internet to find the site of Didier Trono's lab. They distribute free vectors and they have a lentiviral tetracyclin inducible system. I think the name of the vector is plvuttr-Krab, or something like that. You have to not be afraid of cloning, since this system requires quite a few!

free is good (we are quite broke.. it's not a good grant year....). i look it up and it looks like what i need. although i must confess they they have a couple of vectors (plvut, plvct and plvet backbones) and i don't understand the difference between them....
