If we don't use RNAlater, what is the best and simplest method for storing cultured monolayer cells for later RNA extraction preferably without cell harvest.

Without cell harvest? Do you mean you don't want to trypsinise them? Sorry if I seem confused - must be the wine

I used to resuspend my cells in trizol and store them at -80 for RNA extraction at a later date.
