How to count the reading frame from cmv promoter? - (Mar/09/2008 )

I have a construct with have a cmv promoter. I recently replaced a gene with another gene but it seems doesn’t work. I wonder if it got a frame shift, since when I replace the gene all the enzyme I used are blunt end cutting enzyme. I don't know how to check the shift. Help! Pls.

The reading frame starts from the start codon till stop codon. Go over it.

Sometimes the distance between the promoter and gene can influence expression.
