verify my microarray data - need help in real time PCR (Mar/04/2008 )
I plan to do some experiment using real time PCR to verify my microarray result.
May I know normally how many genes people use for real time to verify their microarray result? For example, if i'm using 20,000 genes in my microarray, how many genes (how many percent of the 20k genes) should I use?
Any suggestion (tricks and tips) in choosing the genes for real time?
Or, is there any good paper/protocol that recommended?
Thank you very much!!!!! ^^

We did a microarray a few years back and had 10, 000 genes. We got the bioinformatics department to order them in terms of most differentially expressed (we were comparing KO and WT cell lines). Once we got our huge list, we went through the first 100 or so differentially expressed genes and validated via RT-PCR. That gave us enough data to go on with. But it depends on what you are looking at
