I make MTT assay but I don't understand why we make measurmet using 570nm while background is prepare using 640nm.
How regard this informations when I prepare results??

I think it is mentioned in the product description sheet in the kit.

The 640nm background is to subtract the background reading caused by your 96-well plate. It is recommended to do that.

However, some users do tell me that if you don't have an ELISA reader that can read 640nm, you can skip that as well. Just read 570nm without subtracting any background every time and use the same batch/brand of 96 well plate... you should get a comparable result.
