Polybrene concentration-toxicity - (Feb/23/2008 )

Dear All,

I used a wrong concentration of Polybrene for my retroviral infection of MEFs. I was supposed to use 8ug/ml final concentration of Polybrene in the virus supernatant, but the actual concentration is 40ug/ml, which is 5 times higher. I know polybrene may be toxic to cells at higher concentration, but is 40ug/ml safe? I haven't observed obvious cell death 8 hours after the infection.

You are OK if you haven't seen any serious cells death by 24 to 48 hrs.

This must be your lucky day. Good if there is no toxicity in 2 hrs.

We see toxicity with 8ug/ml in 24 hrs. All of our cells die.
