One of these days - Is this a critical mistake - reverse transcription (Feb/20/2008 )

It's an Oopsday and I've managed to add DTT to my mix BEFORE the annealing step (65 celcius degrees). I've continued the experiment, but I wonder, if there will be any cDNA in my samples. Does DTT interfere with oligod(T) annealing? It can't break hydrogen bonds, can it?

There shouldn't be a problem, as DTT is just for reducing. Keep going, it won't be too many more steps before you see if the reaction has worked (which I'm guessing it willk have).
