Expression vector for Pseudomonas - expression of a 10Kb gene in Pseudomonas aeruginosa (Feb/15/2008 )
Hi all....
My work deals with Pseudomonas aeruginosa which is resistant to most of the antibiotics including ampicillin tetracycline etc. I would like over express an antibiotic gene which is of 10Kb size in P.aeuginosa. Kindly suggest a favourable vector as iam unable to get any vectors. Thanks to all......

10kB is huge -- what antibiotic is it? I work with Staph, so I can appreciate your antibiotic resistance woes!

Thankyou for the reply. Well pseudomonas is resistant to all anitbiotic except gentamycin. The gene we would like to insert into an expression vector produces secondary metabolite belonging to Phenazine group. And this gene is 10Kb size. The secondary metabolite is toxic to E.coli, so we are trying to find which expression vector we can use and transform back into Pseudomonas for over expression. It will be gud of anybody working in this area to give us a suggestion reagrding this problem.
