Storage: cDNA or mRNA - (Feb/13/2008 )
Is it better to store mRNA isolated from brain tissue as mRNA @ -80C or convert into cDNA and store @ -25C? Any major differences in long-term vs. short-term storage? Thanks.

I prefer to make cDNA from fresh mRNA to minimize polyA shortening & degradation. If I'm going to set up an series of PCR from the cDNA over the course of a couple of days, I'll store the cDNA at 4`C or ice for up to a week between uses. Also, I'd recommend avoiding more that 2-3 freeze thaws of either mRNA or cDNA.

anyway, it is not recommended to store RNA for too long. convert it into cDNA and store it as cDNA is always a better choice. as cDNA is more stable compare to mRNA.
