EGFP stable cell line proof - (Feb/11/2008 )

Hi,
I have a question about how to publish a paper on stable cells. I've constructed two stable lines expressing GFP at different levels due to different promoters. Aside from showing the difference in fluorescene ( with a monochromator), what other data should I present in a paper?

Thanks,
joe

Amplification of GFP from your cells and western blot of lysate would be good.

You can show the expression level of EGFP by RT-PCR and Western Blot, it is also possible to use flow cytometry (FACS) to show the intensity of GFP fluorescent signal of the two stable cell lines compared to parent cells.

are your transformed cell lines monoclonal or polyclonal? I wonder if an ideal monoclonal stable transfectant should reflect the polyclonal situation...

so you may compare monoclonal and polyclonal transfectants
